Research Description:
Our lab use Xenopus egg extracts to study structure and function of chromosomes. Highlights of our accomplishments are: to demonstrate that the chromosomal passenger complex (CPC) is required for chromatin-induced spindle assembly; to demonstrate importance of CPC association with chromatin and microtubules to activate Aurora B and spindle assembly; to establish the method to identify proteins that selectively interact with double stranded DNA ends, nucleosomes or specific histone modifications in Xenopus egg extracts; to demonstrate the importance of microtubule dynamics in functional nuclear envelope formation; to establish a method to reconstitute nucleosome functions using recombinant histones and synthetic DNA in histone H3-H4 depleted extracts.
